1.0 Analyses using Propidium Iodide (PI) Staining. 1.1 Pellet fixed parasites for 5 min at 4000 x g and gently resuspend in 0.5 ml of the PI staining solution (90 ml PBS, 10 mg PI, 0.1 ml Triton X-100, 3.7 mg EDTA, adjust volume to 100 ml, protect from light, store at 4^oC). Add 250U RNase cocktail (Ambion Corp., Austin TX) to each sample being stained, mix by pipeting gently and incubate in the dark at room temperature for 30 min.
